> TIGIT Signaling Axis TIGIT (T cell immununo receptor with lg and ITIM domains) is a newly discovered immune checkpoint protein expressed on T cells and Natural Killer cells (NK). TIGIT is an inhibitory receptor which modulates the immune activity by competing with activating receptor CD226 (also known as DNAM-1) for ligand binding. This on and off switch of TIGIT & DNAM-1 parallels that of CTLA4 & CD28, which makes it a promising target for anti-tumor therapy.
CD155 (PVR) and CD112 (PVRL2 or Nectin-2) are co-ligands of TIGIT and DNAM-1, while CD113 (PVRL3 or Nectin-3) is only binding to TIGIT. The affinity between different ligands and receptors vary. The binding between TIGIT and CD155 is more robust than that between TIGIT and CD112 or CD113.

Fig.1 FACS assay shows Biotinylated Human TIGIT, Fc,Avitag (Cat. No. TIT-H82F1) can bind to 293T cell overexpressing human CD155. The concentration of TIGIT is 0.03 μg/mL.
