Product Overview
Vascular endothelial growth factor (VEGF or VEGF-A) is a signaling protein that stimulates the formation of blood vessels (angiogenesis). There are several members of the VEGF family, with VEGF-A being the main and most extensively studied one. VEGF proteins function as homodimers (two identical subunits) linked by disulfide bonds. Each subunit is approximately 40–45 kDa, and the VEGF-A dimer binds to VEGF receptors (VEGFRs) on endothelial cells. Application
This kit is specifically designed for the accurate quantification of human VEGF/VEGF-A dimers in cell culture supernatants, serum, and plasma. The standard used is a recombinant VEGF dimer protein, and the antibodies specifically recognize the VEGF dimer.

Workflow

Storage
Keep the unopened kit stored at 2-8 °C. Avoid using the kit beyond its expiration date. The opened kit should be stored per components table. The shelf life is 30 days from the date of opening.

For each experiment, a standard curve needs to be set for each microplate, and the specific OD value may vary depending on different laboratories, testers, or equipment. The following example data is for reference only. The sample concentration was calculated based on the results of the standard curve. The minimum detectable concentration of VEGF is less than 7 pg/mL.
Sample Values
Serum Sample: Twenty-four healthy serum donors were evaluated for the concentrations of human VEGF in assay.

Intra-Assay Statistics
Ten replicates of each of three samples containing different VEGF concentrations were tested in one assay. Acceptable criteria: CV<10%.

Inter-Assay Statistics
Three samples containing different concentrations of VEGF were tested in independent assays. Acceptable criteria: CV<15%.

Recovery
Recombinant VEGF was spiked into three human serum samples, and then analyzed. The average recovery of VEGF for serum samples is 85.87%.
